Drug intelligence / Profile preview
**[177Lu]Lu-JMF-04** is a novel, peptide-based radiopharmaceutical developed for the targeting and treatment of CXCR4-expressing malignancies. It consists of an optimized analog of EPI-X4, an endogenous peptide antagonist of the C-X-C motif chemokine receptor 4 (CXCR4), conjugated to the chelator DOTA and radiolabeled with the beta-emitting radioisotope Lutetium-177 (177Lu). Developed through a collaboration between Ulm University and the University Hospital Basel, [177Lu]Lu-JMF-04 exhibits high affinity and selectivity for CXCR4. Preclinical evaluation in Jurkat tumor xenografts demonstrated significant tumor accumulation and rapid body clearance, primarily via the kidneys, highlighting its potential as a targeted radioligand therapy. Additionally, its theranostic potential has been demonstrated using its positron-emitting counterpart, [68Ga]Ga-JMF-04, for PET imaging.
